Targeted and Global Metaproteomics

We are working on the development of a protein biomarker capability to detect changes in ocean metabolism on large geographical and temporal scales. This developing workflow involves combining global metaproteomic analyses to discover abundant protein targets, and using multiple reaction monitoring mass spectrometry calibrated with stable isotope labeled peptides to produce quantitative profiles biomarker proteins. We are focused on numerous aspects of the technological development and adaptation, from sampling (see Clio AUV development), extraction optimization, mass spectrometry optimization, accuracy and standardization, and bioinformatic processes.
The deployment of this new method has potential utility in identifying the locations of multiple key biogeochemical processes in the oceans and their responses to anthropogenic and natural perturbations.
