Using untargeted and targeted metabolomics methods, the Kujawinski laboratory has analyzed exudates by Ruegeria pomeroyi, a member of the Roseobacter clade within the α-proteobacteria (Johnson et al., ISME J, 2016). They compare intracellular and extracellular profiles as a function of two growth substrates, propionate, a 3-carbon carboxylic acid and dimethylsulfoniopropionate (DMSP), a quantitatively significant algal osmolyte. The lefthand panel portrays the diffuse oligotrophic ocean with free-living organisms. In contrast, the righthand panel shows dying plankton cells coalescing to form a colloid or particle composed of, and exuding, organic matter. The phytoplankton cells leak DMSP which triggers production of a signaling molecule in R. pomeroyi (orange cells). Once the signaling molecule is present at a sufficient concentration, an array of metabolic shifts occur. (Illustration: Jack Cook, WHOI)

Welcome to the FT-MS Facility at WHOI

The FT-MS Facility of the Woods Hole Oceanographic Institution was established in October 2007 with funding from the National Science Foundation, the Gordon and Betty Moore Foundation, and the WHOI Director of Research. We offer fee-based services to support research in marine and terrestrial biogeochemistry, atmospheric chemistry, and biology. Since its inception, the WHOI FT-MS facility has enabled a suite of investigations into the chemical composition of seawater and freshwater dissolved organic matter, of atmospheric particles, and of biological mixtures.

Our facility offers instrumentation and services to characterize environmental mixtures:

1) Fourier-transform ion cyclotron resonance mass spectrometer (7T LTQ FT-ICR MS, Thermo Scientific): provides ultrahigh resolution and high mass accuracy and tandem MS options for molecular identification

2) Triple quadrupole mass spectrometer (TSQ-MS, Thermo Scientific): for quantitative analysis

3) Targeted and untargeted metabolomics on the TSQ-MS and FT-ICR MS, respectively

3) Versatile ionization modes for a broad chemical nature of samples

4) Liquid chromatography (HPLC and UPLC) for enhanced analyte resolution  and trace quantitative analysis

5) Data processing and analysis services

Marine Metabolomics

Metabolomics is a technique that seeks to measure all of the small organic molecules produced by cells. Because these small molecules are the end-products of multiple levels of metabolic regulation, their concentrations provide a temporal snapshot of the metabolic state or phenotype of an organism at that moment. Metabolomics is a rapidly expanding field that has, until recently, predominantly focused on characterizing human metabolic profiles for biomarkers of disease, but is now expanding to a wide range of organisms and environments. In the oceans, metabolomics is still in its infancy.

The WHOI FT-MS facility and the Kujawinski laboratory have collaborated to develop metabolomics methods that are appropriate for marine systems. These include targeted metabolomics for metabolite quantification and untargeted metabolomics for compound discovery. We offer services for sample analysis, data analysis, and new method development for specific targeted analytes. Please inquire with the facility manager for more information about this capability.
